纖維化 (fibrosis) 是疾病惡性進程中一種病癥，其中更以肝臟纖維化 (liver fibrosis) 與肺纖維化 (lung fibrosis) 最常見。組織纖維化的發生是當組織過度生長和硬化，最終導致過多的細胞外基質 (extracellular matrix, ECM)和膠原蛋白 (collagen) 的堆積而至。
肌纖維母細胞 (myofibroblast) 在纖維化中扮演者重要角色，刺激並活化細胞生成膠原蛋白，最終引發疾病纖維化的發生。

Sirius red 是專門用來染膠原纖維 (fibrillar collagen) 的染劑，這種染劑具有膠原蛋白種類 (collagen type independent) 和檢體物種獨立 (species independent) 的特性，與檢體中的 fibrillar collagen 的 [Gly-X-Y]n 螺旋結構 (helical structure) 位點結合，切片結果呈紅色處即為膠原代表的位置。
研究指出，在肝臟纖維化的檢體中，使用 Sirius red 染劑來分辨肝纖維化評估 (Metavir stage)，染色結果更優於傳統的 trichrome 染劑。
